Should have known this would happen. Bought Mass Effect for PC on launch day too and wouldn't work right out the box. Spent 7 months with EA tech support trying to figure it out before they just gave up and so did I.....weak sauce BioWare....
Fool me once...shame on you....fool me twice...and I am a dumb ass...never again.

Cant blame Microsoft for everything. In my books (and the software engineering world) it's the application developer's responsibility to ensure that their product works on all platforms they claim to support in all testable cases. And when that product is a patch that is supposed to fix crap it had better damn work.

I have the same problem. The game simply will not start . The play button is inoperative. Alll the other buttons on the start menu work, but the game is not playing. I downloaded the patch , as well as the Fury of Shale content, and...nada.
Have you tried the steps listed in the Original Post of this thread?

Dell XPS M1530, purchased 05/08
Vista SP1 32 bit, just updated manually
Intel Core 2 Duo CPU T750 2.20 GHz
3.00 GB RAM
Video card: NVidia GeForce 8600M GT
(Edit: it wouldn't surprise me in the slightest to hear this is actually a Vista issue, if that is, in fact, the case. Vista seems to have been one of the more temperamental OS...)

Following the OP's advice, I did the custom Microsoft Update, selecting every option.
The symptoms matched the OP's. Patch installs. Upon launching from the shortcut, I'm unable to select "Play." When I attempt to start it from the directory, using the EXE, I get a mismatch error message.
I'm chalking it up to a peculiarity of my system/gremlins. While I certainly appreciate the efforts you and others have invested in getting it to run, I'm not going to try again. Seven hours is long enough.

Also wondered why there's no update utility or option in the autolauncher or in-game so we don't have to visit a mirrosite to get the patch. The whole nature of this patch, it's dubious origin, how broken it is . . . something just doesn't feel right here...
P.S Bioware still hasn't responded to my question that i sent 2 days ago regarding this problem...
