I can't decide who to use.. My main character is a Mage.  Then I have Alistar, Leliana and recently I added Shale.  Before Shale I had Morrigan.  I switched for 2 reasons, first because I wanted to try out Shale and second because I thought I could use another tank...  I'm not sure if Shale is a better tank then say Zevran.. I haven't tried the Assassin Elf but he's in my camp..

Well I played on nightmare so my choices were limited if I didn't want to wipe all the time.

I went with me (tank, I suggest alistair for high difficulties, shale for lower difficulties), morrigan CC/dps(with the basic heal spell), wynne (healer with dispel) and leliana (could be anything but I took her for lockpicking)

I found nightmare to be sometimes too easy with this team so I beleive it is very effective, I tried many other combinations but few seemed to equal this one.

On easy/normal just about anything can work but taking at least a mage with a basic heal and a tank will make things easier.

My team is an attacking mage (main caracter), Alister, Leliana and Wynne. It works just fine.

I play a rogue and my other characters are Alistar for tank, Wynn for healing, and I rotate between Morrigan and the murabi for dps. Unfortunately it seems I have to drag Wynn along everywhere unless I want my chars chugging potions even though I'm not very fond of her character

I never took Alistair out of my party on my first character so on the new mage I'm trying I'm going with PC (mage), Leliana, Wynne, Shale.
