I post this only as a followup to a prior dicussion here and because I just spent a frustrating hour or so trying to figure it out. Hopefully if someone comes across the same problem, they'll remember it quickly:
The Plot Assist Marker (V) can only be turned off by the same quest that turned it on.
I discovered this because I was trying to be clever - I figured that at the end of one quest/conversation it would be easiest to just turn the Plot Assist Marker over the "next" guy, then when I got to him, turn it off in his quest. It didn't work: I was simply unable to turn it off no matter what I tried in "his" quest.
There are (at least) two possible solutions:
1) Use the Plot Giver Marker (!) when passing your PC off to the next plot giver.
2) Make both Plot Givers update the same quest.
I chose the second, simply because the plots were related; your mileage may vary.

Plot assist is a per plot system. A single object can be the plot destination for multiple different plots, and each one is turned on and off seperately.
You can set flags from multiple plots in a single conversation however. so if person A gave you the quest, you can set a flag in person B's conversation to disable the plot assist, even if person B has a different quest he gives you. Those can be seperate plots, and don't have to be in any way related.
Typicaly, plot assist should match up with the journal state. So if there is a plot assist telling you to talk to person B, there should be a journal entry as well. When you update the journal entry to say you've talked to B, you're going to have to set a plot flag, so you can use that to disble the plot assist.
