The way they could include romance with characters like Parasini and Shiala is to not make them full on subplots, but more like one night stands. My character in Dragon age had sex a number of times, while poor Shepard is only twice a game, and one of those four times got him choked.

As for your question in the title, the chance that there will be new squad members and new LIs are pretty high, considering they want to accomodate newcomers to the franchise. Or they just keep the current set and give them 2 sets of dialoge, making them as emotionless as a sack of cement, to make ME3 not "overly referential".
I know I'm preaching to the choir, but I would prefer referential... anyone played MGS4? One of my favorite games of all time, in part to the way it wrapped up so many loose ends and didn't give older fans of the series a swift kick to the crotch like so many sequels do.
