Does doing missions with one character on a playthrough transfer to the same squadmate in a 2nd playthrough when you are trying to get the achievement such as the Asari Ally achievement?

No, it doesn't. The real answer here is to only do the very basic requirements to get off the citadel, then go straight to Therum. Then keep her in your active squad for the whole game. According to the wiki it will take 50 assignments to unlock her achievement.

Yeah, and if you do everything in the game(like explore all the uncharted planets), possibly along with PS and BDtS I think you can get 3 of the ally achievements in 1 playthrough. I know you can at least get 2 for a fact...
