I'd played the game mostly unspoiled and fresh (just got into the Mass Effect universe recently, and stayed away from all story discussion until I'd done a play through of each), so imagine my surprise when my femShep was just (I thought) chatting with Garrus like the old friends they are, with Garrus relating his stories about how to relieve stress on a Turian ship, and then the only remaining dialog option for my character was making a pass at him?!?!?! Seriously, it came as a surprise, and I couldn't help but gasp a "WTF??" at that moment.
Same with Jack and Mordin, who both seemed to mistake you talking with them to be flirting with them.
Because of that, I've held off talking to some of the other characters, despite wanting to know them. Thanks to this thread, it seems that I'll be able to talk to Thane and Samara at least, and I really need to spend more time chatting with Legion, it seems.
Is it possible to just chat with Jacob without it sounding flirty? I noticed the first time I chatted with him and asked about him, femShep got a really flirty tone in her voice, so I never pursued that dialog tree again.
So, yeah as far as my friendships in the game go, Tali and Garrus are my closest friends, the ones whose backs I'll always have and whom I know will always have mine. We've been through too much for it to be otherwise. Ash was another good friend in ME1, but I never really connected with Kaiden. Wrex actually saying "Shepard, my friend!" in ME2 was a great moment. Mordin is my best friend of the new characters. Jack is very interesting (not sure if I'd yet call her a friend, but definitely someone I want to know). Grunt hasn't impressed me much, but then I haven't spent much time talking to him. He just didn't really interest me.
I wish we could actually have interactive dialogs with Kasumi and Zaeed; especially Kasumi as I find her far more interesting. But, I'd like to be able to talk back to Zaeed, even if just to say, "Look all your talk of doing this and that? It doesn't impress me. I saved the Citadel from a reaper, so :P".
Anyway, I fully support the notion of getting to know the characters much more deeply without romancing them.

if i can just throw out there all the stuff i would want for me3:
crew banter, teasing, conflict, heck not everyone can get along. CONSEQUENCES....like, hey Shep, I don't really approve of what you did there, I'm gonna leave! and stuff like that.
that would be just awesome. and more talking to everyone. yes i want more rpg elements here, please.

She and Ashley were great friends. Now probably Kasumi, Joker, Garrus, and Liara are her best friends. Tali is very much like a little sister.
There really isn't a squadmate from either game that I dislike. Miranda annoyed me so much at first but now I even like her. Jacob is frustrating just in that he's hard to not romance as a femShep.

Yeah... I'm only on my second playthrough and I tried again to just talk to him, hoping I could do so without sounding like a smarmy idiot. He seems like he'd be a great person to talk to, with his background in the Alliance and his apparent respect for fShep. But less than one full conversation in and out come the cheesy lines...

He was utterly dependable in ME1 and followed me to the end. After getting brought back not from Death's doorstep, but Death's living room, my Shepard felt adrift and alone among enemies. Garrus was the first guy he found from the old days that he could trust. It was a huge weight off my Shepard's shoulders to be with a friend once again.

EDIT:
And we need more Jack in ME3, even if you don't romance her. She doesn't want friendship, that's ok and in-character that you cannot be friends with her in ME2, but don't you think it should change in ME3 (especially if she's loyal)?
My Shepard thinks of Tali the same way. To the point that when I did the Tali loyalty mission, I was ready to draw weapons and start capping those damned admirals.

One of the very best lines of the entire game, period. Just one bit of dialog there and it showed you a lot about both Shepard and Joker.

She's the consummate traditionalist. Not the caricature of traditionalists that certain people like to paint, but the real deal. She's an Asari of the old school, but that doesn't mean she hates aliens, kind of like Ashley is a hard assed Marine of the old school but doesn't hate aliens (doesn't trust them though, and with good reason in many cases). For example, the dominant, modern religion of the Asari is not unlike a type of nature worship. Samara, however, adheres to the old monotheistic faith of the goddess. Whereas most Asari live happy go lucky lives, she adheres to the old code of the Justicar and stands as a champion for her people.
