A blood mage would get mind control options on certain people (or, get in deep sh*t if he tries to do that against a hardened, veteran templar), a ranger might be able to solve a quest in a different manner if it takes place in a forest/nature area., etc. I loved the fact that you got to go into The Fade yourself to confront Connor's demon if you were a mage, and I'd love to see more content that is tailored to your class.
This would make me happy beyond belief. One of my favorite parts of PS:T was stat-based dialogue options; the ability to snap a Dustman's neck before he had time to cry out left me sitting there staring slack-jawed at my computer screen. I guess it could just as easily be rebuilt as class based for DA2.
I hesitate to say it due to the backlash (or perhaps more accurately, wankage) over Mass Effect 2, but I did quite like the Paragon and Renegade interrupt systems. It made dialogue feel more interactive and more dynamic, not just two talking heads standing there and staring at each other. I doubt they'd implement it for DA2 given all the whining over the dialogue wheel, but I'd like to see something like that.
Also, the ability to visually customize armor and weapons: color, style, just little things like that so we don't look like we're running around in patchwork gear. It's a nitpick, but it'd be nice.
