I went to a desert world, first planet I have explored. Ended up in the mako. So I drove and found dead marines and a thresher maw. I killed the thresher. Then I found a probe and a salarian skeleton.
It seems like there is nothing to do on this world. I can't take samples with the mako somehow can I? Was that all I was supposed to do was take a look around and then come back?
ME1: Mako, is there more to it?

Every planet you can land on with the Mako has some sort of mission on it. Go to the map from the start menu and locate a point of interest from there for your objective. You may also use the Journal.
Probes, skeletons, and the like are only a plus. They give you some items, but that's that. Usually you have to get inside a base and do something. Try looking around some more.

Look around, find stuff and minerals, maybe come upon a base full of targets and maybe loot.
AND... defy gravity and pull of some stunts and stuff.

Did you exit the Mako or just ride around in it??
The Dead marines and Thresher sounds like the quest line for the Admiral Khouko(sp) that leads to the Cerebrus quests. You exit the Mako, examine/walk near the marines hear you companions talk about them being his men then go back to him and get an update and later a quest.

Yeah, that definitely sounds like Edolus. Basically examine the bodies of the marines and you're done with that world.
Basically there's a few things to do on each planet.
1) Search for mineral deposits or crashed probes. This gives you money and equipment.
2) Complete gatherer quests. Matriarch Writings, Prothean Data Discs, League of the One Medallions, etc. Usually these are shown on the map as "Anomaly"
3) Missions. These usually involve raiding pirate/mercenary camps and bunkers, destroying Geth outposts, investigating mineshafts, and so on.
Edit: And if you don't feel like like searching every square inch for hidden mineral deposits, I suggest going to the Mass Effect Wiki and looking up each planet. They have nice little maps there.

Yes, be sure to get out of the Mako. I think I explored 3 different planets before I figured out I can get out of the car. Felt really stupid.

Yes do get out of it. You can't make any story progress in it.
I usually open gamebanshee.com when out in the Mako. Its ME1 walk through includes maps of areas with points of interest marked (including unexplored worlds). I use these maps as a road map to make things go faster (otherwise my sometimes OCD behavior would dictate a need for a grid search)

Yup, just to emphasize: You HAVE to get out of the Mako in order to examine/collect stuff, otherwise you won't be able to make any progress.
Since this is apparently Kahuko's first mission, there is no base to go to or anything. Just stop by the dead marines, kill the thresher maw that will appear, then get out of the Mako and examine any one of the marine's bodies and you're done.

Man I am so terrible at this game. I have a tendency to wander out in the open and try to pick everyone off with my gun. Then I realize my whole squad is dead and I am still surrounded. I think I need to remember to get people to cover is the problem. I think theres a button to tell them to hold.
