The powder you get from the person in duskwood. I have gone to my trail 3 time and defended myself all 3 and the powder never came up as a option. How is it used with your trial

You don't do anything with it except carry it to the trial. Once you've gathered the powder and the other evidence, you go to the trial and they modify your chances of winning. When Sand first talks to you, he mentions that the court is examining the evidence, but it's all automatic.
When the trial is over, the three pieces of evidence are supposed to be taken away, but that's a little buggy. Sometimes they go, sometimes they don't. The Powder has never gone away for me - and you can't drop it or get rid of it, except to give it to a different party member; so Sand ends up carrying the Powder for me the rest of the game (rather appropriate, in a way).

First, if you get the Alteration Powder through using your Bluff skill on the dryad, you get an extra point during the Trial (important if you want the Master Orator feat).
Second, it is an empty bag.
Bags hold stuff. Since it doesn't reduce weight, it ought not hold armor & weapons. However, it is great for ammunition or gems (weightless), or potions or scrolls (10 per pound). We adventurers never have enough bags to properly organize the gear for a team. I like having a box for potions (fragile) and a ring for keys (easily lost in the bottom of a bag), but I'll take any sack that comes my way.
Second, it is an empty bag.
