I'd downloaded and deleted Firewalker, because my 360 is a 4GB system, so I've got to be thrifty with what I pick of the DLC, then when I tried starting the game again, it said that I couldn't resume it. Redownloading it fixed the problem, but was this just because I'd apparently saved in a system that only appeared in Firewalker or will I have to keep the Firewalker content on there if I want to utilize that playthrough?

I think you have to have the DLC installed. Or start a new character. But i'm not 100% sure.

If you started a game with a DLC installed then remove that DLC, you won't be able to continue that particular game because it's flagged as having that DLC. So you'll have to eithe reinstall the DLC to continue or start a new game.

What if you delete the DLC and do NG+?

I would guess it would depend on the actual dlc you are referring to.
Certain weapons carry over into NG+, and if those are dlc items then I can see NG+ not working.
I would also guess that if you had obtained any upgrades from dlc missions, and you removed those dlc packs from your storage, then NG+ will not work. The game would have registered that you obtained those from the dlc and would probably require them for the new game.
This is only my speculating though. Perhaps someone can give a more definite answer.
