Personally, I think that this isn't a defense of Earth. Rather, its one of Earth's colonies and Sheppard has the opportunity to somehow stop the Reapers from reaching Earth, (total) IMO.

Mass Effect 2 essentially ends on a cliffhanger (fleet of approaching Reapers).
Technically Bioware could decide to sqeeze a spin-off game or prequel into the middle of the trilogy if they wanted to, but it really only makes sense to do that if you live in Stupid Crazy Land. That would be a little bit like George Lucas making "A New Hope" and "The Empire Strikes Back" and then making "Phantom Menace" before "Return of the Jedi." Fans would be like... "WTF?"
Now it's possible that they could do a totally side-project that is not a PC or console game. Something like a handheld game, or a Facebook game, or some such. But I don't think any such project would get this kind of hype or a cinematic trailer.
There is absolutely nothing about that snippet of a trailer that suggests it is a prequel or spin-off or that it can't be ME3.
So, I say again, they will announce ME3 or I will eat my hat. AND my socks.

Unless Bioware considers this title an integral part of the story for ME3 that they want to tell first. Besides they have already done one minor spinoff, and it'd hardly be revolutionary for them to suddenly change their mind. Baring in mind the books barely even mention Shepard yet still tie in to the main plot point (the Reapers, not Shepard), they could decide to make an expansion of some form to ME2 that does not have Shepard in the main role, yet still ties in to ME3. Besides, Lucas decided to start on number 4, even when he knew he wanted 6 episodes of Star Wars, so he was hardly the master of conventional storytelling.

1. The first Mass Effect teasers (RPG of the Future) came out fairly early 2006. (I scoured the internet looking for old announcement trailers. No easy task.)
2. Mass Effect was released on November 20, 2007, a year and a half after the first teasers. (Source: Mass Effect Wiki)
3. The first Mass Effect 2 teasers (Geth? In MY armor?!) came out early 2009, a little over a year after ME1's release (again, scoured the internet).
4. Mass Effect 2 was released on January 26, 2010 (I was there), a little UNDER a year from the first teasers.
5. Both teasers looked very different from the finished games.
6. It has been said (and here I fail at finding a source, check Kotaku) that Bioware plans on coming out with ME3 faster than the other two.
7. That is totally an M-29 Incisor that Typical Gritty White Space Marine Guy is wielding.
So, assuming that ME3 WILL be announced in December, that pegs ME3's release either late 2011 or early 2012.

Um, Archangel in Mass Effect 2. And Bioware has already confirmed that ME3 will try to be darker, while bringing back some of the lightness and humour from the first game, which they obviously did by not making everything orange.
Also, the Incisor is a new weapon, so it couldn't be set in anything before the events of Mass Effect 2. Unless they did some serious retconning.

The first picture (when scanned) shows the atomic mass (58.45) for Iron.
The second picture (when scanned) shows -128.5 degrees F. The lowest temperature ever recorded on earth.
The first two clues (plus a Mass Effect rifle) point to Aequitas, a planet in Mass Effect 2 known for its "Iron Canyons" and a surface temperature of -128.5F (-89C).
It was also the site of a sidemission, N7: Abandoned Mine in which a Reaper indoctrination device turned a research team into husks & abominations.
Wait, Wait. The tempature was recorded in Vostok, Antarctica. There’s a system in Mass Effect 1 called Vostok. DAMN YOU BIOWARE AND YOUR CONFLICTING INTEL!

Dude, ME3 is supposed to be the last stand against the Reapers. I'm pretty sure all hell will break loose and everyone will look like hell too....
I'm 99% sure it's ME3. To me it's just obvious.

Assassin's Creed was also intended to be a trilogy and it still is... and now we have Brotherhood.
