Ally achievements trigger when a mission and / or assignment is completed except for the collection ones. It maybe possible to get three allies in one play through, but two seems the norm.
For me I got Wrex first, Tali second, and Liara last, the others were muddled in between with the order I earned them.
Edit: I forgot to address your question in the Title...
That one triggers when you complete 15 missions and at least 35 assignments. It should not interfere with getting any other achievements.
Double Edit:

This is a pretty decent checklist with quite a bit of information on it.

You get this if you hack a terminal on Citadel.maxut85 wrote...

thanks very much for the links guys. I'll check them out. And I figured out I had a save from about 5 minutes back where I didn't accept the assignment so I thnk I'm good.
You won't even need to worry about getting assignments, only completing them with squadmates matters to register towards the achievement
Plus it seems to log to your profile not your characters so you could have three shepards and have taken Kaidan and Ash with you on 15 assignments on each one and then trigger once you get the minimum on the four character.
Just remember the game is intended to be played at least three times starting from scratch to even get to level 60 and unlock Insanity.

I just finished a playthrough with Garrus and Liara and I didn't get either ally achievement. I went to Therum first, completed 45 assignments with them and took them through the entirety of Noveria, Feros, Virmire and Ilos. I guess I have to download BDtS and hope the puts me over the edge. My first playthrough I got Kaiden and Tali just fine, I wonder what I did differently?
IIRC the trick was to not do (complete) any assignments BEFORE recruiting them, especially Liara.
So, Spectre - Therum - go back to Citadel, do all side missions - continue the main plot..
You should get the achievements way before Ilos..

That's what I tried to do. I picked up some (but I made sure I didn't complete them) while I didn't have Liara, but Garrus was in the squad basically as soon as possible.

That's weird then, maybe a bug? I do remember getting Liara's ally achievement on a side mission, way before Ilos. Her was the last one I did not have at that time.
edit:
in any case BDtS and PS (if you have it) should help too!

Well, I got Garrus' achievement during BDtS and after I beat the game it showed I had Liara's as well but I never saw the achievement flag pop up. I'm guessing the safest way is to not even activate the assignments until you have the people in your squad.

Hmm... I'm not sure why you experienced that problem, did you count the collection assignments towards the achievements by any chance? Maybe that could account for what you experienced?
I'm on the 360 with the game playing off of the HD, so I'm not sure if there could also be something with playing the game that way or not and how the info is tracked towards the achievement.
My other question is if Pinnacle Station would count properly for Allies, because Shep is solo when you gain EXP after leaving the simulator, I didn't use it when I got Liara's, so no idea, (Liara was the very last Ally achievement I got).
Regarding achievements in general for ME, I've had the most trouble with getting AI Hacking and Neural Shock, the others just take time.

I didn't count the collection assignments nor
Virmire: Kirrahe's team rescued
Virmire: Wrex is convinced
Excluding those I have a total of 48 completed assignments (counting those only on the assignments tab) after having killed Saren part 1 at the Citadel. When I previously played through and didn't get the ally achievements I hadn't done BDtS and had 45 completed assignments. The only remaining ones I want to get are Wrex and Ashley ally and the Insanity playthrough. Then I have to start over on ME 2 achievements
I don't have PS so I can't comment on that one.

Good luck with the ME2 achievements they're much more fun to get.

Your not missing much without Pinnacle, and the wiki said Garrus and Liara take 50 assignments each, Wrex 40, and the other three 45, but I also wonder if there is anything else to it that the wiki does not mention?

They're also much easier to get. I've only done two playthroughs and have all but 2 or 3 of them. A far cry from the first game!
