Can't say that I'm "sick" of it/them (Jack/Samara), however, I haven't played ME2 since July. That's probably helped.
Lately, from browsing through threads that I previously hadn't ever paid attention to, I've come to like other characters more than before, though - namely Ashley.
Who's sick and tired of their favorite character?

For Wrex yes, I don't ask about krogan politics anymore - it seems I can repeat those lines even awoken at 5.00 a.m. For Kaidan - latest of my runs are full renegade and I didn't want him to see these Shepards, so saveedited and now enjoy nearly the same lines by Kimberly Brooks. For Kasumi - downloaded recently, enjoy. For Miranda - I talked with her more and, after hearing about her final thought towards to Cerberus, like her far more.
The only character who has dramatically fallen in my eyes is Thane. Hear about metaphysics, dead wife and long lost son at first time (I haven't played KotOR and hated Sky from JE) was exciting experience, at the 16th...

I do understand how one becomes bored with something they like, though. It's almost akin to overexposure.

I'm not tired of my favorite (Tali) but I'm definitely not hogging her in my playthrough I'm doing now. Infact, when I get from ME1 to ME2 again, I'll be turning down her advance. If I want to savor my Talimance, I'll load up that file

but I can understand if that happens, it depends on the view of the player, so we are all individuals and have our own point of view!

Yeah I am now trying to take as many different characters with me now as possible to try and hear different combinations, and trying to find out which character responds to other characters' comments (like how Miranda responds to Jacob on Omega). Quite awkward sometimes though, and time consuming!
(Angel the character improved drastically once he got out from under Buffy and stopped pining for her, whereas Buffy's whinge factor increased over time. Angel the show really started to take off once they started working for Wolfram and Hart, which is a shame since it is then when they cancelled it, I was especially looking forward to the possible fight between them and the Slayers that was implied to be a possibility in one episode).

That sounds familiar... too much playing, I suppose, and not THAT much variety to it. Good thing is, you focus on other characters and find different things there. ME3 will rectify that sickness, I´m quite sure.
On a sidenote, dusting off your (in general terms) very first Shep might prove difficult, though - with that many Shepards you´re bound to lose the emotional investment for one set of character traits (read: the You-Shepard in your head, the one with the clunky face) that made the first playthrough special.
On my first playthrough I romanced Kaidan and felt terribly crappy after Virmire - good that that kind of desicions were there. Can´t really imagine that I´d feel much when teaming up with him again on the third. *sigh*
Then, after a long while, played ME again, sparing Ash. And, to my horror, it was almost "meh".
On the bright side, you probably know all of the characters so well by the time 3rds out and in your sweaty palms that you really feel as taking some old friends/comrades with you to gang up and spit in the general direction of the Reapers.
