Yes, but with some limitations, as you've discovered. A disclaimer, I don't have a lot of experience playing without internet access, someone else please post to confirm or correct this:
1) You must have a physical disc in your laptop to make a check at start-up of the game. Digital download copies from EA and Steam require an online verification.
2) You cannot use any DLC-- this too is checked online at the start of the game.
3) Any saved games you made with the DLC active (and which contain, say, DLC loot) will also cause problems.
So in essence, if you have the disc, you can only play the game itself, and possibly the Awakening expansion (which would also require a disc). You should be able to "uncheck" the DLC listed in the Downloadable Content screen and then start a new game in your existing installation.

So I don't believe the game is working as intended if you cannot play off-line. I'm pretty sure the disk check on the disc version is the only thing that is supposed to be necessary every time you start the game. Unfortunately, intentions and reality are sometimes different things, particularly where this DLC DRM seems to be concerned.
Cryo84: This is something I keep hammering on just because it happened to fix this for me, but it's a quick thing to check: Does your CD key show up in the Game tab of the DAO Config utility? If not, that's likely the problem, and I can dig up instructions on how to fix that.
You might also try disabling auto-login in the game before pulling the network plug. I remember that for a few ME2 players with off-line play issues, there were only problems if the game tried to log in but failed.

I should also note that if you have a disc version of the game, you can still reinstall it using the EA DM and, after authorizing it once, it should work fine offline. I don't believe you can install the DM version of the game over an existing installation, so you'd have to backup your disc version's saved games and uninstall it first.

The DAOU_UpdateAddinsXML.exe doesn't work, at least not on Windows 7.
I poked around at it and figured out that if you change the "RequiresAuthorization="#" it won't check the sever for autorization every time the game restarts.
The file is located in the users Documents\\Bioware\\dragon age\\settings folder, just do a ctrl+F for RequiresAuthorization and change the following number to "0". this causes the game to not look to the server to play your DLC.
