I'm on my 7th ME2 playthrough, and for the first time I have discovered that after Tali's initial hearing you can go back and speak to Captian Vas Neema and get several conversation topics.  This was also the first playthrough where I heard Garus' "Hospitals aren't fun to fight through" line.

What new things have you found after several playthroughs?

Only 30?

You lucky bastard. I get one every second playthrough.

A secret room where in the last DLC they put one of Mondo's fan fiction into affect.

The one with Tali, Miranda and Jack....

..or maybe I just wished they did that..

Nah 3 1/2 for me and now the trophies are done thats it till a couple of weeks before ME3 as a refresher.

I did discover a lot of new things on insane, mainly different ways to kill things.

Mix and match your squad and there are all sorts of snippets that you won't see with other people in it.

30 is a bit nuts, but you can't argue value for money.

The half is a vanguard I got bored of when it got too easy and since you need a 100% insanity for the trophy it was not much use continuing.

You just go back the way you came in.

Just got to Virmire in ME1 again.

Apparently, you can ram a Colossus with the Mako, then sit on top of it. Bugger can't get up. It's fun.

Playing soccer with the geth on the prothean skyway on Feros is pretty entertaining too.

I always liked hit-and runs in the Mako, sometimes played "Mario" with enemies too. No gold coins though... I have found amusing Zaeed comments though. He's rather chatty in cutscenes involving the Blue Suns.

I'm pretty sure there are still dialogue variations based on the various different outcomes of Tali's loyalty mission that I haven't heard - and I'm just now playing through most of the loyalty missions with Legion because I've decided this time the crew dies.

Noticed I could click on Liara's computer in the admin office on Illium. Said there was no current info, but I wonder if you can use the Cerberus data you can obtain from the N7 mission on Lorek.

I just recently discovered that if you don't persuade the wounded merc on Korlus to lie to his pals and you also don't use the renegade interrupt to stop him from reporting you, a lot of the merc and Jedore radio chatter changes. They're ALL aware of you much earlier, and what's more, they deploy Fenris mechs during the earlier fights.

So there I was running around in the Citadel when all of a sudden I accidentally clicked on the ad-box thing. And there it was. Elcor Hamlet. The greatest thing I've ever seen. Ever.

No. Before you go to the Alarei, when you get the chance to chat up the admirals, you can also head back to the dock and talk to Captain Ysin'Mal again.

Playing soccer with those Husks on that one UNC Colony is more fun. I just wanted to try it out, random cruelty FTW,, attracted one via Sniper round, it went running towards me, I jumped in the Mako, hit the gas, went on full speed and then *WHACK* ... that Husk literally flew over the next mountain, I even had enough time to zoom in on him and tried twice, TWICE to snipe him with my main cannon until I couldn't see him any longer, I think it was still flying...

Nah...that's only for if you give Liara the info on the Shadow Broker before acquiring Thane and/or Samara. Since she leaves after you give it to her (giggity), you wouldn't otherwise be able to get the info you need to find Thane and/or Samara.

All you have to do is pay attention to the percentage next to the name of the systems and star clusters. It tells you how much of those areas you've explored. I never finish the game until all of them say "100%."

They're talking about ME1.  It didn't have exploration percentages.
