I'm guessing it's a choice between the life of the VS + some other stuff, or gaining a significant advantage against the Reapers.

Most likely Earth. It has to be something universal, rather than specific. VS is pretty much meaningless to anyone with the PS3 version. Same with LI's lot's of Shepards don't have them.
Someone also brought up a good point on another thread about it being Shepards humanity. In order to defeat the Reapers he's got to become a "Reaper". That would be very similiar to the Grey Wardens drinking spawn blood to be able to kill the Archdemon.

I think it might be the sacrifice of the Citadel or the Alliance either physically or politically. Try to build a fleet from the various races without them or any political support.

I'm putting five bucks on The Ultimate Sacrifice having precisely squat to do with anything we've seen so far. Another five bucks goes towards this being a self-contained DLC to tide everyone over while we're waiting for the final bridging DLC.
