For me, it was on the Migrant Fleet mission.  The Paragon interrupt to hug Tali when she finds her father's body.  I usually need a tissue for that part:crying:

Yes, I'm sappy.  Deal with it :P

I get a bit tear jerky on overlord, when you beat the AI.

I've only played it once, but I bounced between sadness and fury on that one.  I ended up sending David away from Cerberus.  I don't know if there was a conversation option for Shepard to go uber-renegade and execute the scientist, but I was really tempted.  In the end, I figured David, being autistic, needs all his family and probably wouldn't understand why I killed him.  But I really wanted to shoot that son of a ****.

When escaping the Collector Base reactor overload and then the Normandy flying while the whole thing just spews out a beautiful blast of blue radiation. It is so aweasome I always cry (just a little).

"Shepard! My friend!"

Wrex was the best after that.

That got me the first time, it made me really happy to see an old friend again :D

+ Jack's loyalty mission was very emotional for me, making me realise just how bad Cerberus can be. Also the Archangel mission. Having Femshep shout Garrus when he's lying unconscious on the floor was also a sad moment for me :(

Jack's loyalty mission was emotional for me and her romance was as well. Very poignant and beautiful.

This.

The Tali mission was fun but the Tali-hug didn't trigger any emotional response. But then I'm not a Talimancer either. In fact the most emotional moment of her mission was listening to the final transmission of a Quarian mother to her son, right before she's killed by the Geth.

Overlord hit me like a truck, especially the music playing in the background near the end :(

The Tali-hug really did get me though. The way she was cursing, you could just hear the agony in her voice.  As Okeere said, that part "had weight".

Overlord ending was like "Oh my god..." *SOB* "WHY?!"
Re-uniting with VS and Wrex, and especially Liara was enough to make me drop a tear of joy - and Tali.
When finding Garrus again (:D) and then when he was gargling blood and it zoomed to his eye, I actually screamed and shouted at that moment. NO ONE TOUCHES MY GARRUS! NO ONE! >:(

For me it was being surprised by Archangel being Garrus. Then it was all wash away in embarrassment from the odd way male Shepard got excited by seeing him.

That was a good moment too.  When I started the game, my character felt alone in a sea of enemies.  It was good to have my old bro watching my back again.

When Zaeed throw a inferno grenade at his ex-partner and say "Fry you sonavabiatch".
It's beautiful. I live for these moments of great happiness and celebration.

That wasn't a granade, it was a thermal clip. It landed in a spill of gasoline and ignited, then some barrels blew up.

I always feel sad when I've to choose between Samara and Morinth - Shep is capable to solve almost all issues, but (s)he can't settle a simple family matter :(

Going to town with a Justicar and an Ardat-Yakshi would be awesome

Garrus, Thane, Tali, Miranda, and Jack's loyalty missions. No ranking order.

The Tali loyalty mission was quite full of emotional moments.  Resisting the hug was incredibly difficult when I was playing my "professional" Renegade, then there was "with all due respect I wasn't defending one of your people, I was defending one of mine" and  "I got better... I got you".  I think it works better with Shepard as the father figure than a love interest to be honest, although I suppose both work to some extent.

Mordin singing was pretty great.  The conversation about culture before the song was quite interesting too and helped it hit home how terrible what happened to the Protheans was and even made me feel a little sorry for the Reapers (they seem to be stagnant too).

I quite liked the introduction, the combination of the visuals, the sound and the music is perfect for creating a real impact and I always enjoy seeing it with every new character.

Seeing Garrus again in first playthrough without any spoilers. Had no idea who Archangel was.

Actually, strike that last. It was when Garrus took the missile to the face and we were leaning over him as he threw up a bunch of blood with all that gruesome gurgling, topped off by Zaeed saying "He's not gonna make it..." in an unusually sympathetic tone.
