Tumblr is weird. It's almost like a hybrid of twitter and livejournal (thank god I'm in a place where people actually know what livejournal is. Kids today, you say "livejournal" and they look at you like you just said "usenet." Don't ask what happens when you say usenet.) It combines twitter's transience with livejournal's lack of length restrictions, and makes it kind of complicated and annoying to comment on things or reply to people. Not that I don't use it; I totally use it.
First, you make an account... like you do. To see some posts, you have to either search for tags or follow some people, (much like on twitter or lj). Your home page of tumblr is your "dash," which is where the posts of everyone you follow and all their reblogs show up. You can also make your own original posts from there, naturally.
While Livejournal is all about comments and self-contained discussions, and twitter can be about short, private conversations, the only way to comment on something on tumblr is to "reblog it" with new or added text in the comment field. If you do this, anyone who follows you will see it. You can also reblog something without comment, just to show it to your followers. Finally, you can "like" something by clicking the heart icon; this just favorites it without sharing it with everyone who follows you (it also lets the author know you liked it.)
The main complaint about tumblr (other than the fact that it satisfies all of our novelty addiction with more precision and vigor than pretty much any other service the internet provides, and it can be hideously engrossing for hours on end) is that a lot of people are really terrible about attribution. They'll post a comic or art or quote without linking to the source, so this thing will become a runaway hit on tumblr without the original creator getting any credit. It's super frustrating, especially when you find a funny comic and someone has literally edited out the attribution. Not cool, bro. Whenever you post something on your tumblr, be sure to link back to wherever you found it.

Warning: tumblr likes fenders. A lot. I may only think that because gonebatty was one of the first people I followed on my "Bioware-related-stuff' tumblr, but it's definitely a theme I've noticed.
Tumblr also likes porn. A lot of uncensored porn. So be very, very, very careful about using it at work. (I am saying don't use it at work. Unless you work in a porn.)
