Have you done the Tranquility quest? That opens the quest to recruit Isabela.

It's where you meet and recruit Anders. After completing that, Isabela's recruitment mission opens up once you enter the Hanged Man.

Yep. Doing his recruitment quest is the only way to unlock Izzy's.
