I just started playing DA:2, but I don't plan on using this file long term, since I'm waiting to receive Origins.
Will creating a new game, automatically replace the old character and gameplay? Also, will I lose my unlocks if I start a new game?

Nope. All of your concerns are unfounded, you'll be totally fine there. I've done that multiple times already.
