Can't remember the exact line but when Donnic asks what is going on Isabela just says something about bending Aveline over already. Isablea's dialogue is awesome.
Or when Isabela see Sister Nightngale: "Is there anybody you haven't slept with!"

Also pretty much any time I spend talking to Merrill is amazing as well as when Varric talks to her. The ball of string bit was pretty great.
Generally I've really enjoyed the banter and conversations so far. Another great writing job by BW

Mine too, I think. Hawke has some of the funniest lines in the game, IMO. I got this when romancing Anders:

Hawke can be so weird and out-there. I absolutely love her. The "humorous" Hawke really seems to be living in her own world half the time, and it's great.

Made me laugh quite a bit.

Made me laugh extremely hard.
And I can't find it but the conversation where Varric and Carver insult each other and then ask if they are going for drinks later. That's probably the best.
Hell, anything to do with Varric.
