Why don't the Dragon Age II online profiles work? Do I need to remove the registration and re-enter it? There is an advertisement on the launcher that encourages you to register your game so that your online profile can be active, however it simply doesn't exist.

The Click on "Games" Tab
Then click "Activate Online Profiles" Link.
It might not show immediately, but it should be on. Just go back and play the game. Make sure you have "Upload Character" enable in the DA2 Configuration Tool.

No they don't work now, check the link above, there are many people with this issue. BTW, I clicked activate many times now and I made sure the online features were enabled from the very first time I launched the game.

There's another problem. How to delete a character? I have deleted 2 savegames with 2 different names. But the characters still show up in my profile and there are no options to delete them.

I had set things up on my rig to be able to use the Command Console back in the first week after release. Over the the weekend I discovered that doing so prevents the EA profile from updating. Once I removed the "developer console - enabled" line in the da2launcher.xml file, my portrait, achievements, screenshots, everything uploaded to the EA servers just fine.
I believe that one has to play with the Command Console NOT enabled long enough for the auto-xmit to kick-in, (I've heard that switching gear is one of the triggers for this...) for each character one wants updated.
