I've done all the prerequisites, and she is sitting in front of the Writing Desk, but she will only respond with her banter rather than starting the quest. She still has the exclamation above her head.

I've tried restarting the game, various group composition prior to entering my house and I am pretty much out of ideas.

Is this a bug or is there something I'm missing?

I haven't encountered the problem myself or know what could cause it, but I think you can fix it with the generic companion-problem-solving method:

1. Enable the console and type in "runscript zz_isa_debug", which starts a dialogue.
2. Choose "Plot".
3. Choose "No Rest for the Wicked - Act 3".
4. Choose "Jump to Plot Point".
5. Choose "Quest Start".

Or possibly one of the other options for step 5.

For the record I had the same issue. It seemed like it occurred because when I arrived and she was there the quest "Beware of Dog" had started and she would not start "No Rest for the Wicked" either before finishing "Beware of Dog" or afterwards. I attempted Memodown's suggestion and it ported me next to her but now the quest start "!" was gone. I have a save with only ~30 minutes or so to lose but I would prefer not to do so if anyone has a different suggestion. Its time for me to sleep so I can attempt to solve this tomorrow some time =)
