If I were to start with the new/premade 20 character for Golems of Amgarrak and wanted the Grim Reaper Achievement, which class would be the best to choose, and which spec should I aim for?

Nightmare? I honestly doubt that is possible actually. On hard, well almost the same.
Not only will your char with lvl 20 be very low for the task at hand -- killing the Harvester on nightmare isn't really easy even for many lvl30-ish character builds -- also your companions will be at that low level. Also you would lack all the equipment that make a powerful char. Maybe there are pro-gamers that would manage to, but I think it will be quite a pain.
Easiest run on nightmare I had against the Harvester was with a lvl 35 Mage. I guess a Rogue and/or Archer could be a good pick, for they are very strong in general.

If I were you i would make a mage, Spirit Healer/whatever, probably AW. Points into magic, few into will. Get the SH healing spells, heals, elemental spells do good damage on the Harvester. Respec your rogue mate to be an archer, or you can repsec the other dwarf to be a better tank. It will be very difficult to do, even on Hard, I managed it with a level 12 import but it was very tough. Good luck!

From Awakening onwards, archers actually became outrageously powerful. They way overcompensated for archers being weak in Origins. Be a warrior, make sure you have the archery talent Accuracy and a maxed-out Spirit Warrior spec, and sink your points into Dexterity. The rest is pretty flavour-to-taste. The important points are Accuracy and the Spirit Warrior talents.
That said, powerful as this is, you are still massively handicapped by being level 20 and having no cool gear (that I know if). Perhaps you could do a run through Awakenings just to beef up and kit out a new Warden for the task...?
But yeah, Spirit Healer/Arcane Warrior is probably your other best bet...
