It is directly related to the main quests in Act 2, however, it is a companion quest from Isabela, so I assume you'll have to be up to speed with her before it initiates, plus being far enough along the main story for it to trigger.
That is about all I can say without spoilers or completely backtracking - I can't remember the exact detail of when it triggers.

Riiiight! Now I remember too! It's practically at the end of Act 2, and when you actually engage in the quest there's barely any points of turning back anymore to do side stuff.
