I'm in Act 3 and Isabella is randomly in my house looking at my letters desk with an exclamation mark on top of her head, but when I go to interact with her  there is no cutscene or anything, she just says a random line like "I'm bored". Is this some kind of glitch?

gotta be a glitch...you tried to go out and in the estate?
you have a save that is set short time before it which you can load and then try it again.

The quest "no rest for the wicked" should start at that point (after a cutscene).

There is a workaround for this, though it will skip some dialogue.

Follow these instructions:
#1. Enable the Console with instructions here:  http://dragonage.wik..._(Dragon_Age_II)
#2. Load up your save
#3. Press the ` key. It also has a ~ symbol on it.
#4. type this exactly, then press enter (without quotes): "runscript zz_isa_debug".  This should bring up a conversation menu.
#5. Select Plot
#6. Select No Rest for the Wicked. Act 3.
#7. Select Jump To Plot Point
#8. Select First Velasco Encounter.

This should get you on your way.

The console fix is the only one I've seen. I'm not sure that there's anything you can do on the PS3/360.

Be sure that all the triggers for that quest are filled.

1. You've spoken with Isabela in the Hanged Man.
2. On the Loose has been completed.

Same issues on PC version, also have the broken "who needs rescuing" quest issue mentioned elsewhere. Will try the debug console workaround, thanks!
-edit- debug console worked fine, got and completed quest, although I don't think the final Isabella armor upgrade dropped.  Also, not used to having to type blindly in a debug console (no GUI? or command-line?) but it worked.
Thanks!

You can also try going to your house immediately after talking to Isabella(make sure no one else is going to be in there, so start in your home, go to hanged man to talk to her, go right back home). This worked for me twice so far. Then you don't have to have "On the Loose" finished.
