In terms of old NPCs, seconding the love for Shiala or Gianna Parasini, slightly leaning towards the latter. She's snarky, fun, and seems like she could be quite skilled. Of course, she could technically be dead depending on your playthrough. ;-)

In terms of a new character, I'd like to see some female aliens other than the asari. A female krogan would be REALLY interesting, especially as it could create some possible "angst" over whether it's a good idea to let a possible fertile female who is precious to the krogan risk her life (perhaps she's sick of sitting around the female camp and wants to kick ass like the guys XD)

For new folks, I'd say a Batarian. It'd be nice having a Batarian who isn't an evil arse or a nobody like the guy on Omega who sells stuff.

Hm. The classic number two in a space opera / science fiction background is Commander Riker, of course. A "Chewbacca" type of character might be nice either. Personally I'd like a character who simply shares my view of things, but more focused in biotics than my own Shepard, who is infiltrator.

Actually Kaidan's return would be nice. The problem with that would be the pending romance of my Shepard or not. Even in fiction I kinda find a romantic engagement of the captain of a military vessel with one of their own officers most unappropriate...

"Squad mate" is a limitation of course. Of the characters already existing characters I'd very much like to see Shiala. She already indicated some kind of romantic interest in Shepard, she is a very well trained asari commando who knows how to fight and she's a woman of compassion. On the downside she seems to be not quite mentally stable - both being indoctrinated by Sovereign and consumed by the Thorian left sever emotional scars. Not to mention the fact that she's green now.

A nice funny character would be Han Olar - we didn't have a volous squad mate yet. And since my Mordin is dead I need a replacement science officer.

Not quite as squad mate, but as my personal represantive and spy in the council would be nice for Gianna Parsini. She's not a fighter, so she won't make an actual squad mate in combat situations, but I love her wits, her sly ways and underlying sense of justice and particularly detest for corruption. Not to mention her good looks. I like her attitude, I like her looks, I guess she'd make a good leader... too bad However. And I want her to be forced to wear skirts. ^^

If I think out of the scope of existing characters, a perfect squad mate would be another human woman, with no sexual tension between (Andrea) Shepard and her, who has a mix of Biotic and combat abilities, a knack for command to take Shepard's place when needed and shares most of Shepard's general view on society and the galaxy.A human vanguard, recruited from C-Sec. Actually Miranda is getting quite close, even though her views differ greatly from my Shepard's. Who knows, maybe Oriana becomes squad mate in ME3? However, considering how Mass Effect 2 was concluded, Miranda choosing Shepard of Tim, I would like her to remain my first officer, despite our differences.

Nobody wanted a female Turian? Aside from FINALLY seeing one in-game, it would be great to finally give Garrus a member of his own species to hang out with. Maybe that old flame of his, "Ms. Flexibility" herself? That'd make for a fun few cutscenes.
