Let's say I have finished the game as a paragon and start a new game with the same character and play as renegade... How does it effect a potential import in ME3? Do the decisions from my 1st playthrough matter or the ones from the 2nd playthrough?

You can choose which endgame you want to import to ME3 , so as long you keep them all saved up , don´t forget to backup the autosave once cleared .
It will give you check points like ME1 save games , council saved wrex alive etc .
