So I've recieved the quest from Anora to check out the Alienage. However it is still greyed out on my map, and I can't get there. Am I missing something? Also, if you tell Anora you will back her before the meeting, can you change your mind, or are you locked in?

You need to physically walk through the gates that lead to the alienage to unlock it I believe. You can't go back to the main gate.
And yes you can change your mind. And by then she'll already have played into your dastardly hands, so no worries on retaliation on that.
