So my question is, asides using 'set gender' after selecting a male character, turning to female and keeping all dialogue flags set to male in the game, is there any other way to force a same-gender sub-plot in the game? I ask because i'v always done the Liara sub-plot, however to me an Infiltrator class seems more like a female thing (Call me sexist, I don't care, lol), and i'd prefer to do the Ash sub-plot for once. More or less because my character is going to be a 'closet alien hater'.

You might as well look up ways to mod the game. You're looking to do things the game wasn't originally built to do.

Lol ok. I always play male Shepard, and I always do the Liara subplot, because I'm always disgusted by Ashleys attitude to Aliens. However, my next playthrough I would like to play a female Infiltrator, because assassination style characters seem to me, better played as females.
Using the console command, you can use setgender 0 or setgender 1. If you have a male character, and you use setgender 1, it will change the look // voice of the character to female. However according to wikia, it will not change dialogue flags (Usually stuff that is set for specific reasons, such as keeping male characters having dialogue options with female characters, and vice versa).
Since I don't want to play the game being referred to as "he", when my character is a "she", I am wondering if their is any way to pursue the romance sub-plot with Ash, while using a female character, without using the setgender command.

I'v checked online for mods. I'v seen on forums people talking about the mod, but google is not turning up anything except the same thing, none of which pointing to a mod.

Short of modding the game I don't believe so. Ash wasn't built as a F/F LI and won't offer that kind of relationship to anyone giving off male dialogue flags. Look into modding, I'm sure someone has done it already.
