I don't care for the VS or the FNG Vega. And I couldn't care less if anyone else previously available as a squadmate makes it in. Sadly it is probably (or with some, certainly) the case that they'll appear as non-squadmates, or at a best, temps.

I believe that all LI's will be perm squad-mates plus the confirmed team mates.
If this were to be the real squad makeup then Vega, Ashley/Kaidan, and the Prothean DLC Squad-Mate would replace Zaeed, Kasumi, and Samara. Although I'd expect to see the fore mentioned in some cameo appearance or temporary team style play.

I think the number is going to be ten. The reason I'm guessing two other new squadmates is because I thought I remembered reading a twitter comment about there being 'other new characters' to reveal. If I were choosing there would only be 8, and the final three would be Jack, Grunt and either Miranda or a new female alien.

VS, Garrus, Tali, Liara and Vega are confirmed, right? I assume it will be 8, with DLC character 9. As the ME1 and 2 squads were pretty balanced between combat, tech and biotics, i'd assume that this will be the case again.
Let's say you get 4 squadmates no matter what: Liara, the Virmire Survivor, Vega and Mr(s). Not-yet-known. So you always get a strong biotic, and either a strong fighter or a sentinel. Vega looks more like a combat squadmate, but i'd guess he will be something like a vanguard or infiltrator, so that he isn't a second Ashley. As human biotics are a little bit overused, i'd say short-range-combat-tech-hybrid. To make the guaranteed squad balanced we would need a "vanguard" and a technician. This will be new characters, one of them DLC. This way we would have each combination of tech, combat and biotics once, only lacking the Virmire casuality.
So that everybody has a chance on a soldier and a sentinel, i think Grunt and Miranda have pretty good chances. But with Tali and Garrus, we then have to few biotics compared to tech squaddies. So let's throw in Jack for good measure. What else should she do? 9 characters (10 with DLC) is still less than 12.
So we have 1-2 Infiltrators (Vega, Garrus), 1-2 Engineers (New Guy, Tali), 0-2 Sentinels (Kaidan, Miranda), 1 Adept (Liara), 1 Vanguard (New Guy), 0-1 gal between Adept and Vanguard (Jack) 0-2 Soldiers (Ashley, Grunt). I guess that's as balanced as it gets. The new tech-only char would be DLC.
Everyone who doesn't return was either DLC in 2 (which can be a problem), or has good reasons not to (Legion might have Geth business, Mordin in on Sur'Kesh, Samara might have something Justicary to do, Thane is dieing, Wrex is governing, Jacob...might take a bullett for Shepard. Now that would improve his popularity!)

I think that's very possible that Legion & Mordin aren't squad mates though. Mordin and Legion aren't love interests, most people seem to think (erroneously) that Mordin's close to dying, and Legion is recruited so late in the game in ME2 that you really miss out on a lot with him. Also, Mordin (and probably Legion from the looks of it) didn't have a new outfit in the demo that was shown. It's possible that it simply wasn't ready for the demo, though.

Wrex and Jacob are pretty close to outright being confirmed as not squad mates.

There's some indication that Thane is not a squad mate because of that (paraphrasing) "Thane, Zaeed, and the rest are doing stuff in the galaxy" quote from a magazine. Although it's not really solid because of the inclusion of the "rest" (which means other/the rest of the crew).

Jack's redesign seems to imply being a squad mate, but I also may have read somewhere that all major characters are supposed to get a redesign in some way. There's some doubt cast on that as Mordin & Legion didn't seem to have new outfits in the demo, however.

There's some doubt as to whether Samara is a squad mate since it's possible that they don't want 2 asari (Liara is a squad mate for sure) on the team, especially when they basically fill the same role (biotic specialists). Also, some say that Liara's alternate appearance pack is a nod to Samara (it makes Liara look like a Justicar), so that's another indication that Sam's not a squad mate. There's also the fact that Samara essentially said that she'll kill/confront renegade Shepards if they cross paths again. But, she also said that she'll help paragon/neutral Shepards.

So, with all that in mind, here's my guess:

Maybe Grunt as well to bring the number to 8/9.
