I don't necessarily think we *must* have five squadmates be permanent, but if I had to choose five for that status...

Liara, Virmire Survivor, Garrus, Legion, and Thane.

There are other squadmates I want to see in ME3 even more than some of those five, but I'd almost prefer they *not* be permanent.

Considering BW said that Liara, the VS, Garrus & Tali are technically the confirmed "permanent" squaddies - Im pleased.

Though to be completely honest - I just want Garrus, Tali, & Mordin  And I want Wrex back so badly, but its obvious he's only coming back for a short time. So if I had to pick 5....
Garrus - My one & only true love. I need his badassness around at all times. Plus his puns/comments are priceless.
Tali  - Close friend and someone I see as a lil sister.
Mordin - Trusted friend and advisor. And who else is going to sing to me!? XD
Legion - His time in me2 was WAY to short and I  him.
 
and the last person - Im torn b/w Kasumi [Another close friend to my shep and absolutely hilarious], Thane [he's well...he's thane!] and Grunt [my adopted son]. XP

Tali: My love interest, she's also the tech specialist.
Zaeed: Complete badass & we need Mass Effect's version of a Mandalorian to return!
Grunt: Since Wrex isn't returning as a permanent squadmate, we need a Krogan. Plus, Shepard is Grunt's battlemaster, so I doubt Grunt won't be a squadmate.
Mordin: We need a medic, one who sings makes it all the better.
