Have you ever seen this Mass Effect 2 scene before?If it isn't already the case for you, try to imagine for a moment that your best friend in the world was also of your preferred gender and pretty sexy in a normal kinda way. This is somebody who gets you like nobody else, who can always make you laugh, who never lets you down. Somebody who means more than just some fling, or one of those learning relationships you have before you find the person you're meant to be with. Somebody who always makes some hidden, buried part of you think, "Dammit, why can't I find a guy/girl like you?" because it's always so easy to relate to them. So easy to feel close to them and happy with them. But you never thought of them that way before, so you don't for a very long time.
Then it dawns on you. This sneaking, unbidden thought creeps in: "Wait... why not?" Sure, you think of them as being like a sibling now... but that doesn't quite settle the question. Sometimes, two people are just friends, just sibling-surrogates. Sometimes that's exactly what they should be, what they were meant to be. But just once in a while, it simply hasn't hit either of you yet that what you were both looking for all this time... was right there under your nose all along.
Combine that with having that person as your boss or employee, while working hard on saving the world, and you can see a bond that has been developing a long time.
And, I mean, if your best friend is your best friend, then sure, that's that. But if you had a best friend who happened to also be pretty hot... well, who better to give it a try with than them?
Plus, compare Joker/Shepard to Shepard and anyone else in the series, and you see how unique a romance it would be. This is why I love the pairing, because Joker reminds me of the kind of guy you could actually fall for in real life, whereas Kaidan and Garrus and Thane and most of the others are sort of esoteric choices. Not that I don't like them, I just mean that Joker would be a refreshing change of pace that way.
Also, I've never heard of a romance with a handicapped person in a sci-fi setting. I'd love it if he were an option because it says that a person having a condition doesn't mean he's not man enough for the strongest woman in the galaxy. It's so incredibly much the opposite of shallow.
Also, Seth Green rocks. You've gotta give me that one, if nothing else.

Yeah, Kelly isn't really a romance, just a fling, though I wouldn't mind if they upgraded her. Samara on the other hand, I would LOVE to see become a romance. I'm straight, so that's pure character-love talking. I ship her with a certain kind of Shepard much like I ship Joker with a certain kind of Shepard, and I am dying to roleplay that out. It would be so unique and so intelligent a romance.
I'll understand if they don't do it, but... I love Samara. I want to see if her having a child with Shepard would result in a normal asari. I want her to be happy and have a child that will live and thrive like she hoped so hard her other children could do. I want her daughters to be happy because she's having the baby they can't have. I want sunshine and bunnies and fluffy rainbows in her future! I don't want her life to end with that dream unfulfilled.
I don't even know if I want kids yet, but Samara... she obviously did. I want her dream to come true.
