Ah, full Story Arc. I can imagine a few things that she would need to deal with. Some things relating to her family, others relating to her profession. Being framed for something she did during a mission and needing to clear her name. Or something as simple as needing a rescue from an impossible situation.
As for Romance? Pierce needs more than just a talk then she's ready to jump in the sack. She's not a hedonist or sucker for that kind of thing. She needs trust, faith, devotion and understanding and gives it to those who deserve it. Who've earned it.
As for her relationship with Ashley? I can actually see them getting into a relationship after the SR1 goes - even if it's just for each other's comfort and company. When ME3 rolls around with all three of them talking about what happened while Shepard was dead, I can see... some 'fun' and awkward situations, especially when explaining things to him....
In the gameplay, the Pierce/Williams romance could be triggered by a paragon/renagade persuasion check during a conversation in ME1

Not sure about the renegade, but paragon could go like this. This would be a cutscene literally as soon as the Normandy left the dock after being grounded.
Pierce: Nice to see you're taking an interest in someone.
Shepard: You trying to imply something?
Pierce: Oh, C'mon, Shepard! I have to be blind to not see the writing on the wall! You've got that star-struck look every time you come back from a conversation with Williams!
Shepard: I like her.
Pierce: Jon.
Shepard: Alright, yes! I love her. Alright?
Pierce: Good. Ash is a funky chick, I like her. Although I don't like her Tennyson. Give me Odysseys any day.
Shepard: (
Paragon Dialogue) Look, We're heading into hell.
Pierce: Yeah. Ilos. I know.
Shepard: And, there's a chance that I - we - won't come back.
Pierce: I swear to god, if you say what I think you're about to say. I'm going to punch you.
Shepard: Please, just look after her.
Pierce: 'Look after her' as a friend, or as a close
friend.
Shepard: I don't want to say famous last words, they tend to bite in the ass.
Pierce: So does alluding to them. But whatever, if it puts your mind at rest, then yes. If worse comes to worst, then I'll look after her.
Shepard: That's all I ask.
