How to obtain special powers?

I completed the game once and received two powers. They are AP ammo and Slam. How can I get more in my second play through?

Everytime you complete a loyalty mission and get the loyalty of your crewmate, you should receive access to their special power. Example is Miranda gives the power slam. Once you start a play you will be given the option to pick a single power out of all the ones you have unlocked in loyalty missions. If during the game you find you don't like the power you can go to the upgrades terminal and research the advanced powers(iirc) and then choose a different one from the unlocked powers.

Yes.  Use the research station in the lab for advanced training.

Singularity is only available to Shepard as an Adept

Stasis is available by completing the Lair of the Shadow Broker DLC.

You can check your squadmates (in the squad/level up screen) to see which powers they can use. The 4th is their bonus power which unlocks after completely their respective loyalty missions (Miranda's bonus power is Slam, Jacob's Barrier, Jacks Warp Ammo etc). You only need to unlock those powers once; and on all other (old and new) playthroughs you will be able to use Advanced Training on the Normandy to get it on Shepard.

There's one other thing. When you're going to help Samara to unlock her bonus power (Reave) you also get the option to kill Samara and have her daughter take her place - Morinth has her own bonus power (Dominate). Make sure you save your game before this mission - help Samara (to get access to her Reave), reload, and kill her (to get access to Morinth's Dominate). When you have unlocked both bonus powers, they will be available on all playthroughs like the others.
