Nope. The ony thing that affects how much experience you gain, apart from xp scaling with level, is wether you kill things on foot or in the Mako. 60% penalty for killing from inside the Mako.

Quickest way: Play through main missions and skip dialogue, skip Mako-heavy side missions.

How to get most experience in a playthrough: Do everything you can and always get out of the Mako to kill things (you can soften them up with the Mako first though, there's no XP penalty for that).

It really is that simple.

In a single play through, with the achievements that add to XP gain, you can generally get to level 58 doing absolutely everything (including finding all minerals, asari writings, etc) and killing every enemy on foot. I think my last play through I got to around 580K XP in a single play through. Also, be sure to go through every conversation tree, as codex entries give a small percentage based XP reward, and conversations often yield codex entries.

EDIT: To save time riding around in the Mako (and to make sure you do everything), I suggest using the planet maps found at the wiki so that you can more easily find where things are on any given planet. Also, this checklist can help greatly.

ANOTHER EDIT: Also, be sure to always be able to hack every terminal/container, because each one will give you XP. A comprehensive guide on maximizing XP gain (as well as a couple of infinite XP exploits) can be found at the wiki here.
