Here are mine:

Minsc (Baldur's Gate series) - Because he butt kicks for goodness!!!
Daelan Red Tiger (Neverwinter Nights) - He's ugly, but he's a tank and noble to the core.

Jolee Bindo (SW: Knights of the Old Republic) - Because every aspiring Jedi needs a smartass Master at his side.
Black Whirlwind (Jade Empire) - A big guy and an unapologetic murderer to boot. But you can't help but laugh at him.

Urdnot Wrex (Mass Effect) - He practically wins by default, as he more personality than anyone else on your squad. Still, he's a great character, and has some of the funniest lines I've heard in gaming.

Garrus Vakarian (Mass Effect 2) - What a difference a sequel makes. Went from being the most bland member of your crew to an asskicking rebel. He's also the best romantic interest Bioware has ever made.

Shale (Dragon Age: Origins) - Shades of HK-47 to be sure. But I found this smartass golem to be infinitely funnier. And her backstory was quite tragic and moving.

Aveline Vallen (Dragon Age II) - Wow. Watching the previews, I never imagined her becoming my fave. But what a character Aveline was! Strong, yet quite vulnerable at the same time. And her companion quest was hilarious.

Javik (Mass Effect 3) - There were so many ways Bioware could have gotten this character wrong. Thankfully, they managed to pull it off and create a truly badass companion for Shepard. He's my third favorite behind Wrex and Garrus.
