This may or may not be good news, depending on who you speak to, but there's Kinect support...
Some stuff off the top of my head: class-specific execution moves, weapons with customizable scopes and such, and (based on screenshots and gameplay I've seen) Kaiden, Ashley, and Liara will be squad-mates (whether it's temp or permanent, I'm not sure). Wrex speaks to Shepard over the radio in one mission, so he'll likely make an appearance.

Handheld himself has returned! DoNotIngest's BSN experiences will improve greatly!
Virmire Survivor, Tali, Garrus, Liara & James Vega are confirmed "permanent" squadmates, though someone said "permanent" might be getting redefined a little (Wtf?). Wrex is a temporary squadmate, and Mordin shows up on one major mission for sure, though it's unclear if you'll get him on your team.
You can customize weapons, add many different enhancements to them, there's a high-damage omnitool-blade attack now, you can get stealth kills on weak enemies, you can shoot the pilot of a gigantic Atlas Mech and jump in the cockpit yourself, grenades are back (though not those awesome frisbee grenades, these are standard human grenades), enemies are apparently smarter, and you'll have to recollect your "new" squad, even the members that were always beside you, for some reason or another; even your love interest (better be damned good reasoning on this one).
All I can think of at the moment.
Edit: And apparently there's a huge amount of upgrades you can tweak around. Very nice.

Huzzah for Handheld! Let me say that you haven't missed much (as far as the DA2 forums go). Yep, Ashley is back and has gotten a slight makeover. I don't think the others mentioned the makeover part. She looks a bit more feminine now (or at least has makeup) with longer hair - still looks like Ash, though as far as I can tell.
