Who will get the biggest roles?

Very few of the squad mates got an important role to play in ME1 or ME2. Things might change in ME3 and some squaddies might get an important part in the story. I can think of few,
1. Liara might get the biggest part because she is the shadow broker and is alive in everyone's playthrough.
2. Mordin seems to be some what important in the demo.
3. Miranda got something, but Casey don't say anything because its a spoiler.
4. Tali might have something to do in geth/ quarian conflict.
5. Garrus is squad mate in all 3 games but never got anything important. So BW might change that.

Any ideas?

My money is on Liara and Vega. Liara will be the poster girl, Vega will be the new Jacob. :/

Liara, Ashley or Kaidan, and James (if you're a noob). In that order. If not noob James will take a back seat to prior love interest(s).

Liara and the VS. They were cameo'd so they could survive and play a role in ME3--I have no doubt it will be a big one. Vega will be important, but to a lesser extent since he just seems like a common soldier. He's not as important as the Shadow Broker and Spectre.

Everyone else may play a role, like Mordin and Wrex with the genophage, but it will have to be something we can do without them, since they can die. It may be important overall to the cause and the future of the galaxy, but in terms of gamplay I'm doubtful.

I tend to think this.  Hopefully each will get their due, but I can't see someone who could be dead having a majorly huge plot point hinging on them.  Unless Bioware has some type of black magic... (which I've suspected all along)

Jenkins.

Without joking, I don't know. So far it seems that a lot of squadmates (Liara, Wrex, Mordin, Tali, Legion, possibly Garrus and Miranda/Jacob/Jack in the Cerberus plot) will have a lot of impact in the story.

I would guess Liara and the VS, as they both have to survive until the 3rd game. Not to mention the VS was merely a cameo in ME2, Liara got her own DLC at least.

I would guess Liara, VS, Miranda, and Mordin. They seem to be the most "tied to the plot", even though two of them can be deceased. Although anyone who allows Mordin to die in a playthrough is a heartless individual. You gotta love Mordin.

It'd be nice if Ashley and Kaidan would get some love and Liara would shut up since she got her own freaking DLC that was all about her taking over for the Shadow Broker...which she's now apparently given up scant months after getting it.

Liara and VS as unkillables. Oh, sorry - I mean, new Shadow Broker and second human SpeCTRe.
Tali and Legion as parts of Geth/Quarian conflict
Wrex and Mordin as parts of Genophage story
Miranda because of connection with Cerberus.

He can be dead, so I wouldn't get your hopes up...

None of the characters who could've died in ME/ME2 will have 'game-changing' plot points, though like someone else said they might have a huge impact on the galaxy and individual storylines (I can think of Mordin/Wrex with the genophage, Tali/Legion with the geth/quarians, etc).

Also don't forget that we'll have new characters of some form.

Liara, Miranda, possibly Kaidan ( being a Spectre ) are both "intel" or strategy mates as well as fighting mates , so i can see their importance.

I call Miranda, because yeah okay she can die, but she also has scary big plot armour during the Suicide Mission.. Not to mention, I think a big part of ME3 will focus on Cerberus being the Reaper's lackies, so that's where Miranda kind of ties into the plot heavily.

He's a perma squaddie in all 3 games, he really doesn't need any bigger role.

I'm hoping that Miranda, Legion, Mordin, and Wrex have rather large parts in the game and I think its pretty safe to say they will. They are each apart of a large faction in the game. Miranda - Cerberus. Legion - Quarian/Geth conflic and Mordin and Wrex for the Genophage.

Probably Liara, when she appears before Commander Shepard on the eve of battle offering salvation via biotically-enhanced impregnation.

Realistically? I don't know. I'd really prefer if none of them really stood out from the others, by which I mean 'make them all important in one way or another'. Still, Vega might be nice, if for no other reason than to justify his existence. Why are you here and not <insert favourite ME2 character name here>, huh?
