I don't think the VS will play a large role (unless Ash and Kaidan both end up with pretty much the same lines again...),Liara,Tali,Legion,Wrex and Mordin will probably plot relevant but since all of them (except for Liara) can die they might not have such a big role either and Liara already had her own DLC.
Garrus might take on a more important role but honestly? I think it will be relativ equally divided
Who will get the biggest roles?

Liara, followed by the VS as something plot vital.
After that I think everyone else will all play important supportive roles which will depend on how you acted with them in ME2

I doubt Garrus will take an 'important' plot role, but I wouldn't be surprised if we see him taking on more of a leadership role on the Normandy. I always feel like he's the perfect Tigh to Shepard's Adama. Minus the alcoholism.
Plot wise, Liara will probably be this game's "Illusive Man" in terms of giving out missions and relaying plot specific dialogue. Miranda may not be a squadmate, but she'll definitely be tied to whatever Cerberus is doing. VS is a toss up. They'll be plot specific at the start, probably showing up at the trial. Past that depends solely on how much dialogue they share.
And of course, Wrex, Mordin, Legion, and Tali for their respective races.

For squadmates who have big story roles my money would be on Miranda, Tali, Mordin and Legion all having connections to fairly big story points.

Liara, as beautiful as she is intelligent, with impenetrable plot armour!
The VS might have some importance, but the only one I see being pivotal is Liara, who will probably make Shepard to embrace eternity against his will again or something.

i think miranda will be working along Shep . Depending how you play her, she hands in her resignation to TIM on the Collector Base.
so devs will probably have her working for Shep. I quite see her and Liara advising Shep on how to bring down Cerberus and TIM.
