Are there any blackstone
irregulars quests which REQUIRE you to return to the blackstone
irregulars in Lothering, in order to complete? Or can you just go to any
blackstone irregulars outpost?Someone on the bioware
forums told me that there was at least one blackstone quest which
required you to complete and return to dane's refuge in Lothering, and
told me to make sure to do that BEFORE Lothering is destroyed. When, exactly IS Lothering destroyed by the way? Immediately after completing just ONE treaty quest? Also, for alistair, being a sword and shield tank, which spec should I choose for him, champion, or berserker?

No, there is no Blackstone Irregular quest that requires you to turn in at Lothering. However, there is a Blackstone Irregular quest objective (talking with Patter Gritch in Scraping the Barrel) that is located in the Lothering Chantry.  The Blackstone Irregulars, like the Mages' Collective, is located throughout Ferelden and is part of a larger organization.  A quest from the Denerim Irregulars can be turned in at the Redcliffe Irregulars' turn in.  It doesn't matter.

Lothering is destroyed after a main story quest is completed.  This can be A Paragon of Her Kind, Nature of the Beast, The Urn of Sacred Ashes, Broken Circle, or the quest dealing with saving Redcliffe.

Also, for alistair, being a sword and shield tank, which spec should I choose for him, champion, or berserker?

The Templar talent allows for tanks to fight effectively against mages.  The Champion talent allows for tanks to protect themselves better against melee attacks and offers an AoE stun.  The Berserker talent allows for higher health regeneration and a bonus to damage.  It is all about personal preference.
