Just by looking at them, their snout shape. If its squarish, its a stallion, if its round, its a mare.
And 90% of the characters known on the show are mares. There are stallions too, but they are mainly background ponies.

Stallions tend to be of more neutral or earth colors as well. Whereas the mares tend more towards pastels.
