I have the original DA:O. Could I buy the Ultimate Edition now and have the expansion and all the DLC work with my current saves?

I'm thinking this horse has been beaten several times, but I can't really find a straight answer. All I ever see is "herp derp dlc dun work."

For the PC version, yes, your saves will work fine with the UE. Just make sure that you save your characters folder when you uninstall regular DAO (and DO completely uninstall DAO) before installing the UE.

The Characters folder is in Documents\\BioWare\\DragonAge\\Characters. When you uninstall the game it will give you the option to save those files or delete them as well. I generally make a back-up (copy it and save it elsewhere) just to be safe.
Another note: Any mods you may be using will need to be re-installed as well.
