Arrested Development.
The worst part of that show's cancellation is... I can see why it was cancelled. Don't get me wrong, I love the show, and it's my favourite comedy of all time (as well as in the top three of my favourite television shows, period), but it was incredibly self-referential. Which is fine if you're on HBO, for example, but for a comedy on a major network, it's difficult to do well when a lot of the humour requires you to have watched the previous episodes.
Still. A tragic loss of a fantastic show.

Also, bonus answer - Veronica Mars. Though it never recaptured the magic of the first season, and season three was a bit of a jumbled mess. Still had some great moments, but you could tell they knew they were not long for this world. As I understand it, they finished dead last in the ratings for their timeslot twice in a row, so I have to give the network credit for letting it survive as long as it did.

I can only hope I won't have to add Fringe to this list, though FOX seems to be pretty intent on keeping it around as their 'WE LIKE GOOD TV TOO, GUYS' show.
