This is a tough one and it's hard to narrow it down to one game. I'd say Planescape: Torment was the best written and most memorable. Tie Fighter Vs. X-Wing had the best gameplay. Wing Commander 4 had the best acting. KotOR 2 had the best companions. NWN2 had the best toolset.
I think for the sheer number of hours I put into the game, the user mods, as well as the quality of Mask of the Betrayer I have to go with NWN2.
There can only be one......BEST VIDEO GAME OF ALL TIME

In that case I have to say Dark Age of Camelot: the amazement I felt when seeing Prydwen Keep, Camelot and the Salisbury Plains for the first time, the epicness of Darkness Falls, the sense of belonging both to your guild and to your realm, the amazing RVR system... I could go on and on. If only we could see a DAoC2... sigh, guess I'll have to see how Prime Battle for Dominus will turn out.
If we're talking specifically single player though I have no idea... probably Mass Effect or KotOR2... or Monkey Island... Indy 4, Grim Fandango... The Longest Journey, Ultima IX... who am I kidding? I can't choose!

The Curse of Monkey Island.
Bursting with character and humour, superbly written, visually perfect (it still looks wonderful) with some fantastically thought out puzzles - and some fantastically silly ones - and moments of surreal delight.
It filled me with glee when I was introduced to it as a child and it still fills me with glee today.

there will never be a greatest video game of all time. thing is video games keep on coming out. like there is no greatest movie of all time or greatest book of all time. it's not possible until video games stop being produced or books or movies stop being produced.

Hands down, the first and best video game I ever played in my entire life. From the very first moment, any human being with a soul falls in love with the music by Michael Land. Then you meet the protagonist with the most ridiculous name in history.
And that's only the beginning. The story becomes great, the puzzles and humor make everything fun, makes you feel inventive and brings a smile to your face. The sequels continued to be just as wonderful, but the first Monkey Island will always be my favorite in the series and the best video game I have ever played (and still play).

Final Fantasy IX
Excellent story, simple but effective ability system, plenty of charm and a beautifully crafted world. It was also the FF the series' creator Sakaguchi always wanted to make, and you can see why. Also, I remember how pleased with myself I was when I finally beat Ozma.

I play video games since about 1990 but for me it is actually Mass Effect 1.
I remember it was the only game ever, when, while playing, I thought to myself "this is the best game I have ever played". Combat system, story, pacing, character development, art style everything was just perfect. I didn't really mind the inventory or the repetitive exploration either. Just a great game, kudos BioWare.
Deus Ex is a close second which is why I can hardly wait for Human Revolution.

Most important overall to the Industry has to be Ms. Pac-Man. There were games that came before it that were bringing a wider audience to gaming, but when Ms. Pac-Man came out and made it into the Bars pretty much everyone started seeing Games as legitimate "not nerd-specific" entertainment.
