 I did a poll on this a while ago.  Which Mass Effect characters do you feel should be expanded upon in other media forms such as comics, books, machinamas, etc.  

One idea I had was for Saren and the twenty years he spent with Sovereign between Revelation and ME1 to be expanded upon.  There is really a lot that could be done within that time frame.  Revealing Saren's original reaction to discovering the Reapers, why he didn't go to the Council, how he contacted the geth,  on whether Sovereign told him about the Collectors and their true identity,  when Benzia and her followers caught on,  what he really wanted wtih the frieghter that he had Wrex and the other mercs attack.

I also think Kasumi could do for some more exposure since I feel she has the best personality of all Shepard's crew.  Going into her heists,  the time she spent with Keiji,  how her crimes may connect with other characters within the Mass Effect Universe, etc.

Things I'd want to see-

Miranda's early days with Cerberus. Huge potential to show the reasoning why she is so committed to the goals of Cerberus and the Illusive Man.

Ashley's experience in boot camp. Her first taste of being shunned and dismissed by her superior officers.

Rupert Gardener - losing his family to batarian raiders. It'd be pretty interesting to see how such an everyman could get into Cerberus.

Wrex - clashing with Jarrod after the Rebellions and escaping (maybe some merc missions too)

Ken & Gabby - Tech Academy (who the hell doesn't want to see this?)

Dr Chakwas' earlier days - She's lived a long, adventurous life

Morinth's early life and escape could be interesting. Could find out more about Ardat-Yakshi, Justicars and Asari culture in general, as well as see what kind of crazy sh*t she has done to survive, and how her mother Samara became a justicar. =)

[Nixter Shepard]

Samara- Her whole life story seems interesting
Garrus- His time with his squad and his transformation into Arcgangel
Wrex- Early days before meeting Shep
Joker- Flight School
Jack- After her escape up to her capture
