As in a Tank, 2 DPSers, and a Healer; 2 Tanks, a DPSer, and a Healer; or a Tank, a DPSer, a Supporter, and a Healer, etc.

I'm currently using a Tank (Alistair, soon Shale), a DPSer (myself as a DW Warrior Elf), a Supporter (Leliana), and a Healer (Wynne). I'm debating if to replace Leliana with another Tank (so keeping Shale & Alistair), Ohgren (sp?) for more DPS for Sundering (a Human, an Elf, and a Dwarf Warrior team, all specialised in different Weapons sounds exiting), or Zavran (sp?) for more DPS and keeping Lockpicking shenanigans (but are 2 DW too much?).

Well you asked for the best setup. And best setup in an RPG normally means the single more boring playstyle available.

Well, true enough, but a fun setup in MY opinion (aka, not fact) IS part of a good setup.

With the last two play-through's that I've done I can vouch for the Two Front-Liner Fighters (Melee), one Ranged DPS and one healer or character with the heal spell for the moments that would be useful. I don't necessarily call the front line fighters tanks if only because while the shield makes them more 'effective' as a tank I've noticed that if you have a healer just wearing plate armor in general makes the character a tank..

The Sword and Shield combat is a preference I've noticed, my 2 hander could tank just as well as Shale could (Even Shale had trouble pulling aggro off of him) and could take even more such damage as him. I will say that despite that factor the sword and shield equipped with Blood Dragon armor could 'still' take more damage then him if only because due to his armor everyone that hit him ended up being one damage unless they were anyone not using single or duel wield weapons. (Yes, Archers still hurt Im assuming due to Armor penetration)

That is however the setup 'I' have fun with I would honestly suggest that you take whatever setup that you find you would enjoy and if something doesn't work switch it out. Change and dance around with the options ;)

I have PCs created for about every class. Got an assassin, ranger, reaver, templar, champ, mage, bard. Wish there was another mage or two that could join the party and maybe one less tank/warrior type. This would give another range DPS option other than PC/Leliana/Morrigan. As it is, the PC can be any type but you only have one healer, two range DPS, and a whole buncha melee DPS/Tank types. Pretty limiting on replayability.

As it is, I prefer the standard PC/Alistair/Morrigan/Leliana. I have those the longest and can thus shape them to be what I need from them in my party. Though on my PC as tank runs I might drop Alistair for Zev or Oghren. If I could get Zevran at level 6 he might be able to be made into a decent range DPS/melee DPS hybrid. As it stands getting him at level 10 or 11 he is just too far down the assassin path to make anything but that viable.

I use Sten as my charger, me (warrior) as a defender, Leliana as my ranged support and Wynn as my healer and support.

Also to make a good charger focus on upping there health, and strength. Defender protects everyone so you should have high strength and good cunning and health. I love using this party and makes nightmare alot more fun.

Me (rogue), Tank (Alistair), Wynne (Heals plox) and then it varies. Sometimes Sten, sometimes Dog, or Shale (rarely). I can't stand the Dwarf, and more than one rogue is like bull nipples, useless. Morgan from time to time, but Mages make it too easy, so I tend to roll with just a healer.

At the begging my party had : 1 tank - 1 rogue - 2 mages

But mage can become out of many so quick if the fight take to long and then are usueless.

Now my team is : 1 tank (Shale) - 1 off tank (Alistair) - 1 roque (Zevran) - 1 mage (my Hero)

I just love mod whcich allow you to reset your points :)

2 rogues, a tank, and a mage/healer. Rogues sneak behind enemy lines, tank runs in to grab attention, rogues proceed to shred enemy mages :D fun fun

Either 1 Warrior, 1 Rogue and 2 Mages, or 2 Warriors and 2 Mages.

My first play through I was a mage so I set up morrigan to be healing only had Alistair to tank and Leliana as ranged DPS. That combo worked well but I am having a far easier time playing a melee rogue, shale as tank, leliana as ranged DPS and morrigan as mage DPS (I got the first heal spell for her to heal if someone gets too low) but using pots with the occasional healing works out a lot better since **** dies so much faster.

**Not recommended for Nightmare**

on the first run i had me (mage) alistair/leliana/wynne but with so much aoe and blood magic it was just a case of the other hitting stationary targets.

Latest run is me Assassin/DW, morrigan (with spirit heal) and no primal over start but plenty of ent, leli archer and alistair. with leli and me both with arrow of slaying and max stealth i have to say it is a lot more fun, tho has been known to go a tad monkey from time to time as i try not to control the npc's and rely on their tactics

3 mages and a meat shield. Mages are the most powerful class.

Especially since Wynne and Morrigan have pretty weak banter, all things considered.

The best party to me is player as Arcane Warrior/Blood Mage (too much fun), Morrigan as the healer, Zevran as the rogue and Alistair as the useless third wheel.  PC handles combat, with some assistance from Zev whenever you control a group.  Morrigan heals fine and has some extra CC and Alistair runs around being Alistair.  This also inspires some of the best banter I've yet heard.  Particularly concerning Alistair and Zevran's bet.

You do realise there are already several mods that let you redistrubte your party's stats?

IMO the best party setup is:

Slot 1 = my PC (dalish DPS rogue)
Slot 2 = Leliana (support / ranged)
Slot 3 = a nice, thick blanket (tanking / damage absorption)
Slot 4 = a picnic basket (crowd control)

Hmm, well:

1) Alistair as tank
2) My mage
3) Wynne (heals ftw)
4) Zevran/whoever (Leliana for bard buffs isn't bad)

Since I'm on my rogue playthrough, it's Alistair, Morrigan, Wynne & sometimes Shale or some alternate just because. :P I think I need to go back and play a mage again the third time to do the skills up the way that I want now. I've been missing my CC/AoE abilities (not that backstabbing people isn't fun).

My only advice is to avoid too much melee - it becomes a bit of a pain to keep them alive.

My first playthrough was with myself (Assassin), Alistair, Sten and Morrigan. Sten dropped far too quickly, i nlarge part because I couldn't be bothered to pay attention to him. I could have just as easily used three characters instead. Or I would've been better off with an Archer or another Mage, I think.

In any case for my next run I'm intending to go with a Mage PC, Shale, Leliana and either Morrigan or Wynne to heal. It pains me to force Morrigan into a healing role, but I'm just not so sure about Wynne's personality and mine working.

I have no doubt a warrior and three mages is best, but I'll keep a Rogue along just for the convenience in picking locks, and to add some variety.

My favorite so far is:

1: Allistair as main tank

2: Me as an Arcane Warrior (playing cc/damage role)

3: Gwynn as healer

4: Leliana as ranged dps/combat buffer

Without CC melee becomes too much micro-management for me to enjoy. Pausing the game to issue individual commands is ok unless needed for each step of combat.
