Joker, easily. Though Wrex, Mordin, and Kasumi have some hilarious moments too. As does Legion, without even trying. Garrus isn't especially funny in ME1, but in 2, he's marvellous.

ME1, it was Ashley, hands down.
ME2 is a hard one. Garrus is hilarious if you romance him. Mordin's both funny and frightening at the same time so fairly hard there. But the one who can make me laugh the hardest is Samara for that one ironic line on Tuchanka.

Joker has some funny lines and Mordin too.

all round i think it can't be pinned to one person. because altogether samara is boring, but then you throw in thoso three or four oneliners and it makes up for it. same as with mordin and jacob. (Wait scratch jacob he is all round boring, no offense.)

ME1: Laira especially right when you start to talk to her, she was so socially awkward in those moments

ME2: Mordin, his fast talk and just general over simplification of things, and his logic talk, he is almost like HK-47 from KotOR 1&2

Overall: Joker, just because he is him, enough said

I do chuckle every time I take Garrus and Tali to the Citadel. Tali's comment of I have a shotgun cracks me up.
