After I assigned some squad points to Shepard (to strengthen Incensiary I think, and made him Commando), now my L2 and R2 and Triangle button won't shoot any Powers that I've assigned to those buttons. When I press them in a fight, Shepard just adjusts his gun a bit. Any Help appreciated!!

Guessing...
I noticed that if your squad mates die, sometimes tweaks you've made to them disappear...so, just make your tweaks, then save them, so that if they die, it returns to your new default...my issues had nothing to do with setting assigned buttons...I didn't play that deep, but may still work for you.
Also, playing the craptastic interface of Heavy Rain now, some buttons didn't work if I reloaded the game...thought maybe the game or my controller were toast...fixed it by just turning off the console then restarting it. Problem fixed. So, try reloading, or resetting, or turning off then turning back on the console.
Let me know if that works. I had a post up here re a psychedelic colours glitch...really sucked, but not getting it anymore, which is good.
Ah...reminded of the PC help advice given by the guys on the great tv sitcom The IT Crowd...

Yeah I think it was a glitch, and I've gone to an earlier save and dived back in. I'm hesitant now to use the skill to assign squad points. Just gonna let em be assigned automatically (cuz the glitch -if that's what it was- happened after I messed with my points assignments).

My suggestion would entail that even having squad points automatically assigned would not work if you didn't save afterwards and your squad mates died in a battle...my method worked if I saved my changes. Not sure, but my issue may have been relying on a bug which revived dead squad mates without having to use the medi-gel.
Maybe try assigning points yourself, then saving the changes so that becomes the default.
What is the facepalmer selling? PC? Gosh...the solutions sounds worse than the cure! However, I'm thinking that maybe it would have been better to opt for XBox over PS3...the games I've liked the most on PS3 haven't been exclusive and the ones that have been exclusive I didn't think were that great (Uncharted games are good, not great). If I'd gotten XBox, I could have played Mass Effect 1, and see if Halo is any good.
Oh well. This whole "exclusive" thingo is annoying.
