Well, most enemies have one, be it the head or legs (husks), but no matter where I shoot thresher maws in ME1 and ME2, they seem to take the same damage. Do they have a weak spot at all?

I tried the tongue, the center of the head, and what I think are the eyes, but I noticed no difference. Of course, with their immense health, differences in damage amounts are a bit hard to notice...

As far as I'm aware - they don't.

Grumble, what's the bloody point, then... huge target, and nowhere special to aim?

I have always tried to shoot it in the mouth / tongue. It appeared that it took a little more damage that way. But it is hard to notice. The wiki claims that it is indeed a weak spot, but doesn't say what the difference in damage is.

If I play Infiltrator again, I'll try to see if there's a properly huge difference from a Widow shot on normal diffuculty (+50% on head shots should be more noticeable).

But I've just completed my insanity Infiltrator, and think I'll play something else next. Normal Soldier or Adept, probably, maybe Vanguard or Sentinel.
